Genomic DNA Cloning and Sequence of Dehydroepiandrosterone Sulfotransferase Gene from Rat Liver

Abstract
In this dissertation dehydroepiandrosterone sulfotransferase(DST) gene was isolated from Fischer rat F344 liver and characterized. One of the ten positive clones isolated from genomic DNA (gDNA) library, using published complementary DNA (cDNA) as a probe, was named as L12 and used for the material of this study. The DST gene in L12 was composed of 5 exons, showing more than 90% sequence homology to the cDNA, and has an open reading frame (ORF) coding 285 amino acids, Compared with published SMP-2a which has exon I and exon II, the intron after exon II of SMP-2a was absent in L12. Analysis of aligned cDNA sequence of other STs showes a putative 3¡¯-phosphoadenosine 5¡¯-phosphosulfate (PAPS), a cofactor of DST, binding domain during sulfation reaction.
